A Precise, accurate analytical UV spectroscopic method has been developed for simultaneous estimation of Rotigotine HCl and Rasagiline Mesylate by using first order derivative spectroscopy. Stock solutions of both the drugs were prepared in methanol and standard solutions were prepared with 7.4pH Phosphate buffer saline. Rotigotine HCl 10µg/mL solution and Rasagiline Mesylate 50 µg/mL solution were scanned in the range of 200-800nm and the first order overlain spectra of both drugs to check zero crossing points. Rotigotine HCl shown ZCP at 239nm where Rasagiline Mesylate was showing good absorbance and Rasagiline Mesylate shown at 273 nm where Rotigotine HCl shown measurable absorbance. Six-point calibration curves shown linearity in 2-12µg/mL concentration range for Rotigotine HCl and 10-60µg/mL concentration range for Rasagiline Mesylate. Regression equation obtained for both drugs were used further for determination of concentration in accuracy studies and developed product. In bulk drug Rasagiline Mesylate shown recovery 98.33-100.93% and Rotigotine HCl shown recovery 101-103%. The results obtain from analysis were validated according to ICH guidelines and were estimated at up to mark. Therefor developed method can be used to determine both drugs in bulk and any dosage form.
