ANALYTICAL METHOD DEVELOPMENT FOR SIMULTANEOUS DETERMINATION OF CIPROFLOXACIN HCL AND OLOPATADINE HCL IN BULK DRUG AND FORMULATION BY RP-HPLC METHOD.

Researchers developed a reverse phase- high performance liquid chromatography method for simultaneous estimation of bulk drugs and formulation. The chromatographic separation of drugs and formulation was achieved on C18 Agilent Zorbax (250 x 4.6 mm, 5µm) column using Acetonitrile: 0.1 % TFA Water (40:60, V/V). The 10 l volume was injected with a flow rate of 1mL/min, and peaks were detected. The retention time for ciprofloxacin HCL and Olopatadine HCl was 2.92 and 5.10 minutes. The method was linear in the concentration ranges of 56-80 μg/mL for Ciprofloxacin HCL and 24-36 μg/mL for olopatadine HCl, with regression coefficient correlation values of 0.9999 and 0.9996, respectively. The % relative standard deviation of precision, accuracy, and ruggedness was below 2 %. Forced degradation studies were performed under oxidative degradation (30% H2O2), acid degradation (0.2N HCl), base degradation (0.1N NaOH), dry Heat degradation (70°C for 5 h), photolytic degradation (254 nm for 5 h). The proposed method was simple, accurate, and precise. Therefore, it can be applied for the routine quantification and degradation of drugs and formulation.
