ABSTRACT

The aim of the present study was to apply combined mixture design to optimize extraction solvent composition to enhance filtration volume and recovery of assay test sample with ethyl cellulose coated tablets. Solvent composition, centrifugation time and additional filtration step were evaluated as variables. Filtrate volume through Whatman™ No: 1 filter paper followed by 0.45 micron PVDF membrane filter were selected as responses. Results indicated that higher organic phase in solvent composition, extended centrifugation time and inclusion of additional filtration step enhanced filtrate volume and assay recovery. Conclusively, design of experiments as an effective tool for optimization of extraction solvent in assay method was established and validated. The proposed method has adequate reproducibility and proved to be accurate for estimation of tramadol HCl in ethyl cellulose coated extended release tablets.
