Catharanthus roseus (Apocynaceae), commonly known as periwinkle, is a perennial plant that is mostly found in southern Asia. A simple, precise, and accurate high-performance thin-layer chromatography method for quantitative estimation of an alkaloid, vincamine from methanolic extract of Vinca leaves was developed. The chromatographic separation was achieved by using a suitable mobile phase of chloroform: acetone: formic acid [5:1:0.5, v/v/v] for which detection and quantification of the method were achieved by densitometric method at 222 nm. This method showed good separation of vincamine in the extract with an Rf value of 0.33±0.02. The regression analysis data indicated a good linear relationship for the calibration plots for vincamine in the range of 200-1000 ng/spot and the correlation coefficient (R2) was 0.9957. According to the International Conference on Harmonisation (ICH) Q2 (R1) guideline, the accuracy and reliability of the proposed method were determined by evaluating various validation parameters.
