OPTIMIZATION AND CHARACTERIZATION OF FAST DISSOLVING TABLETS OF CANDESARTAN CILEXETIL PREPARED FROM SPHERICAL AGGLOMERATES

The aim of present study was to prepare fast dissolving tablet of antihypertensive drug (Candesartan Cilexetil). The research work focused on the improve solubility and in-vitro dissolution of drug using spherical agglomeration technique. Spherical agglomerates of Candesartan Cilexetil were prepared using PVP K-30 as polymer and dichloromethane as bridging liquid. Prepared powder blend was evaluated for different pre-compression parameters like solubility, compressibility index, hausners ratio, flow property etc. and for post compression parameters like hardness, friability, wetting time, water absorption ratio, disintegration time, in-vitro dissolution study etc. The results of solubility of prepared agglomerates were found 0.15 to 0.91 mg/ml and it was observed higher than the pure drug 0.00071 mg/ml. In- vitro-drug release studies showed that 95.47 % of drug was released from F3 formulation. From the results it was revealed that the prepared fast dissolving tablet using agglomeration technique might be used to enhance bioavailability of Candesartan Cilexetil to augment acute and chronic hypertension therapy.
