LC Method for Simultaneous Estimation of Brexpiprazole and Sertraline HCl

Post-traumatic stress disorder (PTSD) which is also called as “Shell shock” and “Combat fatigue” is a mental health condition that may occur in people who have experienced or witnessed a traumatic event such as a natural disaster, a serious accident, a terrorist act, war/combat, or rape or who have been threatened with death, sexual violence or serious injury. A newer combination, named Brexpiprazole and Sertraline HCl indicated for treating post-traumatic stress disorder was selected for strategic Analysis in synthetic mixture with reliable, sensitive and accurate RP-HPLC method. The chromatographic development was carried out on Thermo Scientific C18 (250 mm x 4.6 mm, 5 μm particle size) column as stationary phase and Acetonitrile: Methanol: (55:45:2.5, v/v) pH 7 of mobile phase adjusted with the help of 1% triethyl amine (TEA). Effective separation was achieved by injecting 10µl of the standard solution into column using a mobile phase composition and isocratic elution have been done at a flow rate of 1 ml/min. Developed method was validated according to ICH guidelines. The common detection wavelength selected for Brexpiprazole and Sertraline HCl was 214 nm. The retention time value of drug was found to be 3.62 min for Brexpiprazole and 4.5 min for Sertraline HCl. The method was found to be linear over a range of 0.3-5 µg/ml for Brexpiprazole and 25-150 µg/ml for Sertraline HCl. The percentage (%) recovery was found to be 98.82-99.39% for Brexpiprazole and 98.95-101.1 % for Sertraline HCl. The developed RP-HPLC method was highly precise, specific, sensitive, and accurate. Hence, the method has the ability to use for regular analysis for quantification of Brexpiprazole and Sertraline HCl simultaneously in bulk and synthetic mixture both qualitatively as well as quantitatively.
