STABILITY–INDICATING REVERSED–PHASE HIGH–PERFORMANCE THIN–LAYER CHROMATOGRAPHY FOR ESTIMATION OF MUPIROCIN CALCIUM IN BULK AND MARKETED FORMULATION

Mupirocin calcium is a calcium salt of a substance having antibacterial activity produced by growth of pseudomonas fluroscence. A sensitive and selective stability-indicating reversed- phase high -performance thin-layer chromatography (RP-HPTLC) method is suggested for the determination of Mupirocin calcium. Chromatography was performed on aluminum-backed silica gel 60 RP-18 F254 S plates with 1, 4dioxane: water (3: 2 v/v) as mobile phase. Densitometric scanning was performed at 226 nm. System gave compact bands for Mupirocin calcium (Rf 0.63 ± 0.02). Linear regression analysis data for calibration plots discovered a good linear relationship with r2 > 0.9989 in working concentration range 2000-12000 ng per band. The limits of detection and quantification were 26.62 ng and 80.67 ng, respectively. Stress testing of Mupirocin calcium was done according to ICH guidelines in order to validate the stability-indicating power of analytical procedures.
