DETECTION AND QUANTIFICATION OF A NOVEL INVESTIGATIONAL ANTIHYPERTENSIVE COMBINATION COMPRISING ROSUVASTATIN CALCIUM, TELMISARTAN AND AMLODIPINE BESYLATE BY DUAL DETECTION MODE HIGH PERFORMANCE THIN LAYER CHROMATOGRAPHY

A sensitive and precise high performance thin layer chromatographic (HPTLC) method has been developed for the simultaneous estimation of rosuvastatin calcium, telmisartan and amlodipine besylate in synthetic mixture. The HPTLC method employed aluminum plate coated with silica gel 60 F254 as the stationary phase and mixture of ethyl acetate, toluene, methanol, triethylamine in ratio of 6:2:2:0.1, V/V as the mobile phase. All the components were well resolved under optimized chromatographic condition with Rf value of 0.63, 0.74, 0.31 for rosuvastatin calcium, telmisartan and amlodipine besylate, respectively. Quantitative computation was performed in the absorbance mode at 238nm for rosuvastatin calcium and telmisartan and in fluorescence mode at 366nm for amlodipine besylate. Linearity of component was observed between 400-2000, 1600-8000 and 200-1000 µg.band-1 of rosuvastatin calcium, telmisartan and amlodipine besylate, respectively. The method was successfully validated as per the International Conference on Harmonisation (ICH) guideline and application of same for the estimation of these three drugs in the simulated mixture.
